چکیده

Suicide gene therapy for cancer treatment proposed by Moolten [1] started more than 25 years ago and has gained momentum with little variations in the original technology. At the current statistics, worldwide 1550 clinical trials of gene therapy have been reported (http://www.wiley.co.uk/genmed/clinical) among them 7% using suicide gene therapy. HSV-TK is the most well characterized suicide gene used for cancer therapy and in other diseases without inducing significant systemic toxicity [2] [3]. Chemotherapeutic drugs used for cancer therapy are problematic because they do not discriminate in their mode of action. Currently available drugs in the market are not cancer specific so functional concentration level in tumors cannot be reached without off-target toxicity level. This is specifically true for solid tumors where vascularization is poor and necrotic at the center of the tumor due to low oxygen and nutrient supply [4][5]. However, when the drug is activated by locally enzymatic reactions in a timely fashion, the metabolite is toxic for the tissue. The most widely used suicide genes are Herpes Simplex Virus-1 Thymidine Kinase (HSV-TK), and Cytosine Deaminase (CD) from the virus Herpes simplex or the bacterium Escherichia coli respectively [6][7]. Prior research has focused on the mechanism of initiation and development of tumors. It starts with the mutation of 1 or several genes, then gradually attains more genetic mutation and genomic instability during the evolution of tumor/cancer. Almost all the genes fall in 2 categories – oncogenes (derived from proto-oncogene) and tumor suppressor genes. There are many ways to treat various tumors ranging from benign to metastatic. Among them surgery followed by chemotherapeutic drug treatments is the widely used method of choice that damages the DNA and renders the cells apoptotic. Additionally, tumors can be cured by more subtle ways by restoring the function of tumor suppressor genes or disabling oncogenes, to prime the immune cells to act and down-regulate angiogenesis and metastatic activities. Solid tumors can be treated by intra-tumor delivery of suicide genes. The protein product of these genes catalyzes the formation of highly toxic metabolites following the application of some lesser toxic prodrugs. This leads to apoptosis or programmed cell death of the treated cells [8]. The apoptotic cells invite immune cells that further clear the tumor zone by phagocytosis. Suicide gene therapy is also known as Gene Directed Enzyme/Prodrug therapy (GDEPT) or as Gene Prodrug Activation Therapy (GPAT). GDEPT can either take the form of CBT (cell

PCR detection of thymidine kinase gen of latent herpes simplex Virus type 1 in mice trigeminal ganglia

Herpes simplex virus type 1 establishes a latent infection in the peripheral nervous system following primary infection. During latent infection, virus genome exhibit limited transcription, with the HSV LATs consistently detected in latency infected ganaglia. Following ocular infection viral latency develops in the trigeminal ganglia. In this study PCR has been used for detection of HSV-1 nuc...

Comparative Molecular PCR-RFLP Study of Native Herpes Simplex Virus Type 1 (HSV-1) with KOS Strain

Background: Recent research on several DNA fragments covering open reading frames (ORF) 1-37 shows a new genetic marker in ORF 6 which is specific for differentiating wild-type varicella-zoster virus (VZV) strains from Oka varicella vaccine strain. On the other hand, herpes simplex virus (HSV) genome analysis by restriction enzymes is used to differentiate types one and two of the virus and eve...

In vitro Interaction of HSV-1 ORF P with Both Thymidine Kinase (TK) and an Unidentified Cellular Protein

Herpes simplex virus type-1 (HSV-1) is a neurotropic pathogen of humans that establishes latent infection in the sensory ganglia innervating the site of primary infection. A number of genes control HSV-1 pathogenicity and latency. Open reading frame P (ORF P) is one of these genes that might have a role in latency and pathogenesis. A complication in the analysis of the role of ORF P in the HSV-...

A dual function fusion protein of Herpes simplex virus type 1 thymidine kinase and firefly luciferase for noninvasive in vivo imaging of gene therapy in malignant glioma

BACKGROUND: Suicide gene therapy employing the prodrug activating system Herpes simplex virus type 1 thymidine kinase (HSV-TK)/ ganciclovir (GCV) has proven to be effective in killing experimental brain tumors. In contrast, glioma patients treated with HSV-TK/ GCV did not show significant treatment benefit, most likely due to insufficient transgene delivery to tumor cells. Therefore, this study...

OBJECTIVE The purpose of the study was to explore the anti-tumor effect of ultrasound -targeted microbubble destruction mediated herpes simplex virus thymidine kinase (HSV-TK) suicide gene system on mice hepatoma. METHODS Forty mice were randomly divided into four groups after the models of subcutaneous transplantation tumors were established: (1) PBS; (2) HSV-TK (3) HSV-TK+ ultrasound (HSV-T...

Enhanced tumor cell killing in the presence of ganciclovir by herpes simplex virus type 1 vector-directed coexpression of human tumor necrosis factor-alpha and herpes simplex virus thymidine kinase.

Past studies have documented the promise of herpes simplex virus type 1 (HSV-1) thymidine kinase (TK) suicide gene therapy as a potential antitumor treatment. HSV-TK converts the pro-drug ganciclovir (GCV) into a toxic nucleotide analogue, the incorporation of which into cellular DNA blocks cell proliferation. In this report, we have examined the hypothesis that the effectiveness of HSV-TK suic...
